| Resumo | Objetivou-se avaliar o efeito dos diluentes à base de água de coco em pó (ACP-101c®) e TRIS adicionado em diferentes concentrações da gema de ovo de galinha d’angola (Numida meleagris). Foram colhidos 15 ejaculados de cinco caprinos da raça Anglo Nubiana, semanalmente no período de cinco meses, com auxílio de uma vagina artificial. Em cada colheira, os ejaculados com motilidade massal (≥ 3), motilidade progressiva (≥ 80%) e vigor (≥ 3) foram selecionados. Em seguida, os ejaculados foram reunidos em um pool e divididos em 12 grupos e alíquotas iguais de 0,5 mL de sêmen e diluídas nos seguintes Grupos Controles – (GC1 TRIS, com adição 2,5% da gema de ovo de galinha Gallus gallus domesticus GOGD), (GC2 Grupo ACP-101c®, com adição 2,5% da gema de ovo de galinha Gallus gallus domesticus GOGD) e Grupos Experimentais GE, com a adição da gema de ovo de galinha d’Angola (Numida meleagris) – (TRIS 2,5%GONM; TRIS 5%GONM; TRIS 10%GOMN; TRIS 15% GOMN; TRIS 20%GONM) e (ACP-101c® 2,5%GONM; ACP-101c® 5%GOMN; ACP-101c®10%GOMN; ACP-101c® 15%GOMN; ACP-101c® 20% GONM). Posteriormente, foram envasadas em palhetas francesas de 0,25 mL e congeladas com auxílio do aparelho TK3000® e armazenadas em nitrogênio líquido (-196°C). Após sete dias, as amostras foram descongeladas e avaliadas quanto à cinética espermática utilizando Computer Assisted Sperm Analyser (CASA), a integridade de membranas, a morfologia espermática, a análise de fragmentação do DNA espermático pelo teste OpenComet, análise ultraestrutural por microscopia eletrônica de transmissão (MET) e análise bromatológica da gema de ovo de Numida meleagris (GONM). Foram obtidas as médias e desvios-padrão e procedida à análise de variância (ANOVA) pelo teste PLSD de Fisher para os parâmetros espermáticos motilidade, morfologia, análises da viabilidade espermáticas e a cinética espermática (CASA); e pelo teste de Kruskal-Wallis para o parâmetro de vigor espermático. Nas análises dos grupos contendo GOMN pelo sistema CASA observou-se que os parâmetros MT, MP, VCL, VSL, VAP, STR, BCF e ALH obteve-se resultados semelhantes comparados aos grupos contendo GOGD (P>0,05). Os parâmetros espermáticos dos grupos contendo GOGD e GONM foram avaliados pós diluição e pós-descongelação e pôde-se observar que a MT e vigor foi mantidas análogas. A integridade de todas membranas e acrossomos avaliados não sofreram influência do tratamento (P>0,05) nem do diluente empregado para criopreservação (P>0,05). A morfologia espermática foi igualmente preservada pelas diferentes concentrações de GOGD e GONM nos diluentes de ACP® e TRIS, sem diferenças significativas estatísticas (P>0,05). No teste OpenComet, foi possível observar uma boa viabilidade espermática da GOGD 2,5%, GONM 2,5%, GONM 5% nos grupos TRIS e ACP®. Já na avaliação da porcentagem de fragmentação de DNA espermático não houveram diferenças significativas (P>0,05). Na avaliação por MET não houve ações danosas nas cabeças dos espermatozoides, com membranas íntegras e acrossoma totalmente preservado nos grupos GONM e GOGD. Na análise da GONM verificou-se valores de proteína bruta de 9,37%, pH de 6,99 e lipídeos de 34,56%. A GONM pode ser utilizada como substituto da GOGD proporcionando uma ação protetora das membranas espermáticas, mantendo os parâmetros espermáticos e protegendo o DNA espermático. | Abstract | The objective of this study was to evaluate the effect of extenders based on powdered coconut water (ACP-101c®) and TRIS added at different concentrations of guinea hen egg yolk (Numida meleagris). Fifteen ejaculates were collected from five Anglo Nubian goats, weekly over a period of five months, with the aid of an artificial vagina. In each harvest, ejaculates with mass motility (≥ 3), progressive motility (≥ 80%) and vigor (≥ 3) were selected. Then, the ejaculates were collected in a pool and divided into 12 groups and equal aliquots of 0.5 mL of semen and diluted in the following Control Groups - (GC1 TRIS, with 2.5% addition of the egg yolk of Gallus gallus hen domesticus GOGD), (GC2 Group ACP-101c®, with 2.5% addition of hen`s egg yolk Gallus gallus domesticus GOGD) and Experimental Groups GE, with addition of hen`s egg yolk (Numida meleagris) – (TRIS 2.5%GONM; TRIS 5%GONM; TRIS 10%GOMN; TRIS 15% GOMN; TRIS 20%GONM) and (ACP-101c® 2.5%GONM; ACP-101c® 5%GOMN; ACP -101c®10%GOMN; ACP-101c® 15%GOMN; ACP-101c® 20% GONM). Subsequently, they were packaged in 0.25 mL French straws and frozen with the aid of the TK3000® device and stored in liquid nitrogen (-196°C). After seven days, the samples were thawed and evaluated for sperm kinetics using Computer Assisted Sperm Analyzer (CASA), membrane integrity, sperm morphology, sperm DNA fragmentation analysis by the OpenComet test, ultrastructural analysis by transmission electron microscopy (MET) and bromatological analysis of the egg yolk of Numida meleagris (GONM). Means and standard deviations were obtained and analysis of variance (ANOVA) was performed using Fisher`s PLSD test for sperm parameters motility, morphology, sperm viability analysis and sperm kinetics (CASA); and by the Kruskal-Wallis test for the parameter of spermatic vigor. In the analyzes of the groups containing GOMN by the CASA system, it was observed that the parameters MT, MP, VCL, VSL, VAP, STR, BCF and ALH obtained similar results compared to the groups containing GOGD (P>0.05). The sperm parameters of the groups containing GOGD and GONM were evaluated post-dilution and post-thawing and it could be observed that the MT and vigor were kept similar. The integrity of all membranes and acrosomes evaluated was not influenced by the treatment (P>0.05) or the diluent used for cryopreservation (P>0.05). Sperm morphology was equally preserved by the different concentrations of GOGD and GONM in ACP® and TRIS extenders, with no statistically significant differences (P>0.05). In the OpenComet test, it was possible to observe a good sperm viability of GOGD 2.5%, GONM 2.5%, GONM 5% in the TRIS and ACP® groups. In the evaluation of the percentage of spermatic DNA fragmentation, there were no significant differences (P>0.05). In the evaluation by MET there were no harmful actions on the sperm heads, with intact membranes and a totally preserved acrosome in the GONM and GOGD groups. The GONM analysis showed crude protein values of 9.37%, pH of 6.99 and lipids of 34.56%. GONM can be used as a substitute for GOGD providing a protective action of sperm membranes, maintaining sperm parameters and protecting sperm DNA. Keywords: Caprine. Semen. Cryoprotectant.
